A complete system for rapid, high-level expression via direct transfection of insect cells.
The focus on high-throughput (HT) expression screening to rapidly identify the appropriate expression system for hundreds or thousands of individual targets in parallel has traditionally not involved insect expression systems. The several weeks that are required to create, titer, and maintain viral recombinants makes conventional baculovirus systems too complex and time-consuming for HT processing. For HT purposes, researchers have been limited to cloning open reading frames into bacterial expression vectors to make recombinants that are used to direct target protein expression in E. coli.
Insect cells provide an alternative system for expressing proteins that require certain post-translational modifications to retain solubility and activity. Insect vectors that rely on AcNVP immediate early promoters to drive expression need no baculovirus component, so the time required from transfection to expression is measured in days, not weeks (Loomis 2003).
HT screening of multiple targets in insect cells The Novagen InsectDirect™ System is a complete system that allows for HT screening of multiple targets in insect cells. Included in the InsectDirect System are the following: • pIEx™ series of transient expression vectors for rapid, high-level protein expression in insect cells while eliminating the need to create recombinant baculovirus for protein expression • pBiEx™ series of multisystem expression vectors designed to allow rapid characterization/expression of target genes in both E. coli and insect cells • Insect GeneJuice?Transfection Reagent for high-efficiency transfection of Sf9 cells with minimal toxicity • Insect RoboPop™ Ni-NTA Purification Kit for automation-compatible His•Tag?fusion protein purification |
