Kapa Taq enzyme has been used for the multiplex PCR amplification of exons of genes involved in cancer: BRCA1, BRCA2, MLH1, MSH2 and MSH6. Amplifications have been performed starting from genomic DNA purified from whole blood with EDTA.
As it can be seen in figure 1, multiplex amplification of various exons as well as individually has been performed without any problem. Kapa Taq enzyme specificity and sensitivity is clearly seen using amplification protocols optimized with other commercially available enzymes and without any further optimization (0.5µL Taq in 25µL final reaction volume), showing even higher amplification and better band pattern that with other enzymes.
